It contains a mixture of amylose and amylopectin and is present in the form of granules varying in size from 1 to\u00a0~ 100 \u00b5m. Inside the granule the starch molecules are arranged into concentric layers around a central point called hilum, starting point for the accumulation of amylose and amylopectin<\/span><\/p><p style=\"font-size: 14px; font-style: normal; font-weight: 400; margin-top: 15px; margin-bottom: 15px; line-height: 25.2px; color: #404040; font-family: arial;\"><span style=\"color: #003366;\"><span style=\"font-weight: bold;\">Material required<br \/><\/span>Watch glasses<\/span><br \/><span style=\"color: #003366;\">Thin Razor blade<\/span><br \/><span style=\"color: #003366;\">Brush to collect sections.<\/span><br \/><span style=\"color: #003366;\">Lugol: Iodine\/Potassium iodide solution<\/span><\/p><p style=\"font-size: 14px; font-style: normal; font-weight: 400; margin-top: 15px; margin-bottom: 15px; line-height: 25.2px; color: #404040; font-family: arial;\"><span style=\"color: #003366;\"><span style=\"font-weight: bold;\">Preparation of Lugol<\/span><\/span><br \/><span style=\"color: #003366;\">2% Potassium iodide (KI) and 0,2 % diiode (I<span style=\"position: relative; font-size: 10.5px; line-height: 0; vertical-align: baseline; bottom: -0.25em;\">2<\/span>)<\/span><\/p><p style=\"font-size: 14px; font-style: normal; font-weight: 400; margin-top: 15px; margin-bottom: 15px; line-height: 25.2px; color: #404040; font-family: arial;\"><span style=\"color: #003366;\"><span style=\"font-weight: bold;\">Protocol<\/span><\/span><br \/><span style=\"color: #003366;\">Sections as thin as possible are made with a thin and soft razor blade in a piece of potato chair.<\/span><br \/><span style=\"color: #003366;\">The sections are directly transferred with a paint brush into a watch glass containing water. They are then immersed into a drop of Lugol solution for 1 min and directly collected on a microscope slide in a drop of water. They are covered delicately with a coverslip and observed with a light microscope.
