This is a prerequisite before ultrathin sections, and it allows (1) to estimate quality of sample preparation at the light microscope; (2) to select part of the sample to be cut and examined with the TEM. Indeed, size of ultrathin sections has to correspond to the standard size of EM grid that is 3 mm I diameter.These semithin sections are stained and examined with a light microscope.<\/p><p style=\"margin-top: 15px; margin-bottom: 15px; line-height: 25.2px; color: #404040; font-family: arial; font-size: 14px; font-style: normal; font-weight: 400;\">Two examples are shown on this page with semithin sections through (1) a primary Zea mays root tip; (2) apical part of an undetermined nodule induced by NGR234 on\u00a0<em>Leucaena leucocephela<\/em>\u00a0root. Electron micrographs of ultrathin sections are also presented.<\/p><p style=\"margin-top: 15px; margin-bottom: 15px; line-height: 25.2px; color: #404040; font-family: arial; font-size: 14px; font-style: normal; font-weight: 400;\" align=\"center\"><span style=\"color: #000080;\"><strong>First illustration:\u00a0<\/strong><strong>cross sections (<strong>1<\/strong><strong>\u03bc<\/strong><strong>m)\u00a0<\/strong>through a\u00a0<em>Zea mays<\/em>\u00a0root tip<\/strong><\/span><\/p><p style=\"margin-top: 15px; margin-bottom: 15px; line-height: 25.2px; color: #404040; font-family: arial; font-size: 14px; font-style: normal; font-weight: 400;\">Below on the left: cross section (1\u03bcm thick) made with a diamond knife histo (Diatome) and stained with <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=3151\">methylene blue &#8211; basic fuchsin.<\/a> Cell walls and starch grains are stained pink (see detail in the middle), cytoplasm and the nuclei are stained blue.<br \/>Observation and Imaging : \u00a0Nikon Eclipse 80i microscope.<br \/>We then select a part of sample for realization of ultrathin sections (80 nm thick). In this example ultrathin sections have been stained with\u00a0<a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=578\">PATAg<\/a>\u00a0(see picture on the right) to reveal starch at the ultrastructural level (figure on the right). The starch grains appear as electron dense globules (ga) and are contained in amyloplast
