They involve sectioning very different plant samples: flowers, siliques, stems, roots, seeds, root nodule. The sections were made in (1) fresh material with either a <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=282\">razor blade<\/a> or a <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=601\">hand microtome<\/a> or with a <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=601\">vibratome;<\/a> (2) frozen samples, with a cryostat; (3) samples fixed and included un different media: paraffin, technovit 7100, epoxy resins (Epon, Spurr) and acrylic resins (LR White) using a <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=601\">rotary microtome<\/a>.<br \/>The thickness of the sections varies according to the technique, from 100 \u00b5m (freehand and vibratome sections) to 1 \u00b5m (sections in resins). The fixation and inclusion methods vary, of course, depending on the objective.<br \/>According to the type of sections, protocols for staining and for cell study have been developed and were used routinely. They are listed here.<\/p><ul style=\"margin-bottom: 10px; color: #404040; font-family: arial; font-style: normal; font-weight: 400;\"><li>Routine topographic staining: carmine &#8211; iodine green, <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=379\">hematoxylin<\/a>, hemalun eosin, toluidine blue, safranin FAST green, safranin- alcian blue, basic fuchsin &amp; astra blue, naphthol yellow, methylene blue \u2013 basic fuchsin.<\/li><li>Histochemical staining to demonstrate various cellular constituents: DNA (<a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=418\">Feulgen<\/a>, <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=409\">DAPI<\/a>, Acridine Orange, Gomori), polysaccharides (<a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=400\">PAS<\/a>, <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=426\">calcofluor<\/a>, propidium iodide), applied to paraffin sections and staining of starch with <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=334\">lugol<\/a> applied to fresh sections.<\/li><li>Localization of peroxidase activity on fresh sections (vibratome) or in whole organs with different substrates: DAB (diaminobenzidine), TMB, chloronaphthol, syringaldazine.<\/li><li>Immunolocalization on paraffin sections and freezing sections by fluorescence (FITC, rhodamine, AlexaFluor).<\/li><li>Localization in situ of enzymatic activities on frozen sections: peroxidases (different substrates), dehydrogenases, phosphatases.<\/li><li>Localization of gene expression by in situ hybridization on paraffin sections (non-radioactive method, DIG, RNA probes).<\/li><li>Localization of GUS expression on Technovit sections.<\/li><\/ul><p style=\"margin-top: 15px; margin-bottom: 15px; line-height: 25.2px; color: #404040; font-family: arial; font-style: normal; font-weight: 400; text-align: justify;\"><strong>Microscopes used<\/strong>:\u00a0 <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=1861\">Leica DMI R2<\/a>; Leica Laborlux S; <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=1873\">Nikon Eclipse 80i<\/a> equipped with digital CCDs cameras.<\/p><p style=\"margin-top: 15px; margin-bottom: 15px; line-height: 25.2px; color: #404040; font-family: arial; font-size: 14px; font-style: normal; font-weight: 400; text-align: center;\"><span class=\"bodyPublications10px\" style=\"font-size: medium; color: #003366;\"><span class=\"Style3\"><strong>Confocal Microscopy<\/strong><\/span><\/span><\/p><p style=\"margin-top: 15px; margin-bottom: 15px; line-height: 25.2px; color: #404040; font-family: arial; font-style: normal; font-weight: 400;\">&#8211; Detection in situ of Activated oxygen forms in roots of young Arabidopsis plants using a fluorescent indicator e.g. \u00a0fluorescent HPF\u00a0(hydroxyphenyl fluorescein).<br \/>&#8211; Preparation of Arabidopsis roots according to the mPS-PI (pseudoschiff Propidium iodide) staining protocol. This technique is perfectly appropriote to the architectural cellular study in roots and embryos of Arabidopsis.<br \/>&#8211; Localization of cellular compounds by fluorescence: DAPI for DNA, calcofluor for polysaccharides (see above histochemical staining).<br \/><span style=\"color: #000080;\"><strong>Microscopes<\/strong><\/span>\u00a0:\u00a0confocal Leica SP2 at the\u00a0<strong><a style=\"background-image: initial; background-position: initial; background-size: initial; background-repeat: initial; background-attachment: initial; background-origin: initial; background-clip: initial; text-decoration-line: none;\" href=\"http:\/\/bioimaging.unige.ch\/\">photonic bioimaging center<\/a><\/strong> of Science Faculty \u2013 Geneva University.<\/p><p style=\"margin-top: 15px; margin-bottom: 15px; line-height: 25.2px; color: #404040; font-family: arial; font-style: normal; font-weight: 400; text-align: center;\"><a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=1360\"><span style=\"color: #003366; font-size: medium;\"><strong><span style=\"text-decoration-line: underline;\">Transmission Electron Microscopy<\/span><\/strong><\/span><\/a><\/p><p style=\"margin-top: 15px; margin-bottom: 15px; line-height: 25.2px; color: #404040; font-family: arial; font-style: normal; font-weight: 400;\">&#8211; Chemical fixation of various plant samples (tissues, organs, cellular fractions) and their inclusion in resins (Epon; Spurr; LR White) for cytological studies, ultrastructural observations and immunolocalizations.<br \/>&#8211; <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=1681\">Ultramicrotomy<\/a>: ultrathin sections 60 \u2013 90 nm thick using ultramicrotomes, Reichert OMU2 and Leica Ultracut UCT.<br \/>&#8211; Production of support films on grids (Formvar, Collodion).<br \/>&#8211; Classical staining of ultrathin sections with Reynold\u2019s lead citrate and uranyl acetate.<br \/>&#8211; Functional investigations: cytochemical staining of polysaccharides using <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=578\">PATAg procedure<\/a> and of <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=1692\">enzymes<\/a> (peroxidases, phosphatases, ATPAse); immunostaining with gold for the localization of pectins, peroxidases, membrane proteins.<br \/>&#8211; Freezing of root and shoot meristems which are then fractured at low temperature with the objective to produce a replica of their surface. The technique is that of <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=1588\">freeze fracture.<\/a><br \/>&#8211; Protocols for the preparation of various plant samples have been developed and are made available (see Protocols).<br \/>A prerequisite for making ultra-thin sections is most often essential e.g., the realization of <a href=\"https:\/\/michelecrevecoeur.ch\/?page_id=1324\">semithin sections<\/a> 1 \u00b5m thick and their staining. They make possible to obtain an image of the sample, biological or not, by scanning its surface with a probe or tip.
